培养条件:35~37℃  22~24小时培养
判定方法:黄色葡萄球菌形成淡蓝色的集落。

利用显色酶基质可以在24小时内培养鉴别金黄色葡萄球菌。

(1)本培养基,是添加了显色酶基质的培养基,在35~37℃培养22~24小时后可以鉴别金黄色葡萄球菌,生长的菌落都显色,所以能简单明了的判断。
革兰氏阴性杆菌,肠球菌,酵母样真菌的发育被抑制。
(2)使用方法:培养基表面充分使干燥之后使用。
待检样本直接接种,或增菌培养后涂布接种,35~37℃培养22~24小时。
※请严守培养温度和培养时间。
(3)判定方法:黄色葡萄球菌形成淡蓝色的集落。
        芽孢杆菌(Bachillus sp. )也有发育并形成薄淡蓝色菌落的时候。产生的是编平状无光泽的菌落,请用革兰氏染色等进行鉴别。
金黄色葡萄球菌的确定,需要实施凝固酶试验等的性状试验,请做同定操作。

能简单•准确的判断大肠菌群。

(1) 根据显色酶基质(X-GAL)的配方,能有效、准确的鉴别大肠菌群。

    显色酶基质X-GAL被大肠菌群特异性产生的β-乳糖苷酶分解,生成碱性磷酸酯酶。碱性磷酸酯酶氧化缩合后生成蓝色色素,大肠菌群形成蓝色菌落。
(2) 大肠菌群以外的菌,被抑制发育,即使发育也不显色。
(3) 大肠菌群的发育支持能力出色,对于损伤菌的检测也有作用。

(4) 计数:大肠菌群:蓝色(蓝~蓝紫)的菌落

(5) 使用方法:在44.3g培养基中加精制水1000mL后加温溶解,以121℃15分种高压蒸气灭菌。在培养皿中注入15-20mL,35~37℃混释或平板培养20±2小时。

(6) 培养基成分:

2、本培养基可用作老鼠和人的白血病细胞的初级细胞培养和菌株细胞的后续培养,此外还可用于包括KATO-Ⅲ细胞在内的各种人体癌细胞的培养。
使用方法:
把10.2g本产品溶解于蒸馏水中,使液体总量达到1000mL。等培养基完全溶解后在121℃条件下高压蒸气消毒15分钟。经过高压蒸气消毒后冷却至室温状态。然后在密封状态下向培养基中加入适量的经过高压蒸气消毒的10%碳酸氢钠,(当加入18~30mL时,CO2气体含量下降至5%以下、在温度为37℃时pH值为7.1~7.4)。如果不是马上使用,请使用密封垫圈将其密封并保存在低温(2~10℃)之处。在使用前应另外加入经过过滤消毒的L-谷酰胺0.292g。使用前请根据不同目的添加适量血清。
贮藏方法、使用期限:
请把本培养基粉末用密封垫圈密封保存在干燥低温(2~10℃)之处。使用期限为一年。

Directions
Dissolve 10.2 g of the medium in distilled water and adjust the volume to 1,000mL. Sterilize by autoclaving at 121°C for 15 minutes. Cool the sterilized medium to room temperature and add a proper amount of sterile 10% sodium bicarbonate. (pH will be 7.1-7.4 at 37°C, in an atmosphere of 5% CO2 in air by adding 18-30 mL of sodium bicarbonate solution.) Aseptically add 0.3 g Lglutamine and the desire amount or serum.

Summary
RPMI 1640 is a medium for suspension culture investigated by George E. Moore, former director of Roswell Park Memorial Institute. Up to now, such media were not autoclavable because of decomposition or deterioration of the constituents, so that sterilization had been performed by filtration using a membrane filter. RPMI 1640 Medium “Nissui” 2 is an autoclavablepowdered medium specially manufactured to overcome the difficulties associated with filtersterilized media, such as contamination by Mycoplasma. This medium has growth promoting effect to primary cultures of mouse and human leukemia cells, various kinds of cell lines and KATO-III cells.
Storage and Expiration
Close tightly and store at 2-5°C in a dark and dry place. May be stored up to one (1) year.

Directions

使用方法:
将11.4g的本产品加热溶解后进行冷却,将pH值调整到7.1±0.1,使用蒸馏水将容量稀释到100ml。将其注入试管中后,添加试料,然后加入蒸馏水,达到2倍量。
在121℃下进行5分钟高压蒸汽灭菌后,进行菌接种,然后在30℃下培养16~24小时。酸滴定时,培养72小时。
使用菌株:Streptococcus faecalis”R”ATCC8043。
