类器官(或细胞)的冻存和复苏

需选用生长状态良好的类器官(或细胞)进行冻存实验。 

类器官(或细胞)的冻存 

1.       离心后的类器官(或细胞)中加入预冷的(2-8℃Organoid Cryopreservation Medium(类器官冻存液),吹打混匀后迅速转移至细胞冻存管中(冻存管内细胞悬液体积应大于0.5mL)。

注意:每1mL冻存液推荐冻存1×103-1×107个细胞或对应细胞量的类器官。

2.       将冻存管放入细胞冻存程序降温盒内(降温盒须提前平衡至室温或4℃),随后立即将降温盒放入-80 ℃超低温冰箱中;也可将冻存管进行人工梯度降温处理,如4℃ 静置10分钟,-20℃保存1小时,-80℃保存过夜。

3.       次日或12小时后将冻存管于低温条件下(不高于-70℃,推荐使用干冰或原降温盒)转移至液氮中长期保存。 

类器官(或细胞)的复苏

 4.       提前在37℃条件下预热类器官(或细胞)所需的基础培养基。

5.       37℃的水浴中快速解冻细胞冻存管,当冻存管内冻存物仅剩些许冰渣残留时立即停止水浴并及时转移至洁净操作台。

6.       将冻存悬液转移至离心管中,缓缓加入5-10倍体积的预热的基础培养基,轻轻混匀。

7.       将上步骤所获类器官(或细胞)悬液进行离心(水平离心转子,150-300g3分钟),弃上清,再次加入基础培养基重悬类器官(或细胞)沉淀。

8. 将上步骤所获类器官(或细胞)悬液进行离心(水平离心转子,150-300g3分钟),弃上清后所获类器官(或细胞)可用于后续类器官(或细胞)的培养。

YPD系列培养基属于Rich medium,也可以称为完全培养基。YPD Medium 由精选的蛋白胨、酵母提取物和葡萄糖按适当比例混合组成;YPDA培养基是在YPD培养基中添加了适量的硫酸腺嘌呤。YPD AgarYPDA Agar分别在YPDYPDA基础上加入了Agar,作为固体培养基倒板用。YPD系列培养基用于酿酒酵母(Saccharomyces cerevisiae)的常规培养。

Coolaber推出YPDA MediumPM2011-10×0.5L)和YPDA Agar MediumPM2021-10×0.5L)铝箔袋独立包装(0.5L/pouch),配液过程非常方便。不需要单独称量各组分,也不需要调整pH值。使用时仅需将袋中培养基倒入三角瓶或培养瓶中,加入500mlddH2O,灭菌即可。

有些酵母培养为何要选择YPDA培养基?

YPDA培养基就是在YPD培养基的基础上添加了适量的硫酸腺嘌呤,硫酸腺嘌呤可以防止含有ADE1ADE2突变等位基因的酵母菌株(如Y2HGoldAH109Y187)在长期培养后变成粉红色或红色。

使用方法:

Rich Liquid Media (Broth)

1. 50克的YPD或者YPDA1升的去离子水中溶解。

2. pH 值在6.0±0.2范围内,无需调整pH121℃高压灭菌15分钟。

3. 避光、室温条件下储存灭菌培养基。

Rich Plating Media with Agar

1. 70克的YPD Agar或者YPDA Agar1升的去离子水中溶解(琼脂在高压灭菌后才能溶解)。

2. pH 值在6.0±0.2范围内,无需调整pH121℃高压灭菌15分钟。

3. 冷却到50℃把培养基倒入平板中,室温使其凝固。封口膜封好后倒置保存在4℃冰箱。

为什么我做的平板不凝?

自行添加Agar,务必选用优质AgarCA1331),添加量为20 g/L。固体培养基不凝固可能是由于pH值偏低或者灭菌时间过长(也会降低pH值);水质偏酸也会使得培养基偏酸,建议灭菌前调整YPD系列培养基的pH6.5121℃,高压灭菌15min,及时从灭菌锅内取出培养基。
