Vector Laboratories affinity-purified antibodies are of unmatched quality. These antibodies are prepared using proprietary immunization schedules that produce high affinity antibodies. The antibodies are then purified by affinity chromatography, and cross-reactivities that are likely to interfere with specific labeling are removed by solid phase adsorption techniques. The biotinylated secondary antibodies are conjugated to ensure the maximum degree of labeling without compromising the specificity or affinity of the antibody. 

With some exceptions the recommended dilution for most applications is 1:200.

Technical Information

The horse anti-rabbit Ig antibodies are prepared by hyperimmunizing animals in a manner that produces high affinity antibodies. These are then purified by an affinity chromatography procedure designed to remove any low affinity antibodies which may be present. Cross-reactivities that are likely to interfere with specific labeling are removed by solid-phase adsorption techniques. The final product is then subjected to rigorous quality control assays including immunodiffusion, solid-phase enzyme immunoassays, gel electrophoresis and solid-phase binding assays. In preparing the labeled antibodies, great care is taken to ensure the maximum degree of labeling with no alteration in the specificity and affinity of the antibody. The labeled antibody then undergoes a further series of quality control assays, including immunohistochemical analysis.

DyLight® fluorescent dyes are direct alternatives to traditional fluorophores such as fluorescein (FITC) and rhodamine. The excitation and emission spectra parallel that of other commercially available fluorescent reagents allowing for easy substitution into an existing protocol without requiring any further instrumentation or filter sets.  

DyLight dyes offer a number of potential advantages including greater photostability and brighter fluorescence.  DyLight dyes are completely stable over a pH range of 4-9 making them compatible with many aqueous-based buffers and diluents. The DyLight dyes can be applied as single labels or in combination with other DyLight dyes and fluorophores as part of a multiple immunofluorescent antigen staining methodology.  The Dylight dyes currently offered are DyLight 488 (green), DyLight 549 (orange), DyLight 594 (red), and DyLight 649 (far red).

Technical Information

The anti-mouse Ig antibodies are prepared by hyperimmunizing animals in a manner that produces high affinity antibodies. These are then purified by an affinity chromatography procedure designed to remove any low affinity antibodies which may be present. Cross-reactivities that are likely to interfere with specific labeling are removed by solid-phase adsorption techniques. The final product is then subjected to rigorous quality control assays including immunodiffusion, solid-phase enzyme immunoassays, gel electrophoresis and solid-phase binding assays. In preparing the labeled antibodies, great care is taken to ensure the maximum degree of labeling with no alteration in the specificity and affinity of the antibody. The labeled antibody then undergoes a further series of quality control assays, including immunohistochemical analysis. 
 
DyLight® fluorescent dyes are direct alternatives to traditional fluorophores such as fluorescein (FITC) and rhodamine. The excitation and emission spectra parallel that of other commercially available fluorescent reagents allowing for easy substitution into an existing protocol without requiring any further instrumentation or filter sets.  

DyLight® dyes offer a number of potential advantages including greater photostability and brighter fluorescence.  DyLight® dyes are completely stable over a pH range of 4-9 making them compatible with many aqueous-based buffers and diluents. The DyLight® dyes can be applied as single labels or in combination with other DyLight® dyes and fluorophores as part of a multiple immunofluorescent antigen staining methodology.  The Dylight® dyes currently offered are DyLight® 488 (green), DyLight® 549 (orange), DyLight® 594 (red), and DyLight® 649 (far red).

Description

This IgG preparation is intended for use as a control for primary antibodies made in mouse. This antibody has been purified from pooled serum of healthy adult animals and contains a spectrum of the IgG subclasses present in serum. The control antibody should be applied to the tissue section at the same concentration as the primary antibody to indicate whether staining is specific for the antigen or is nonspecific adsorption of primary antibody to tissue sites.

Could you please describe what would be an appropriate negative control for IHC that can be used to help validate tissue staining results?

A commonly used negative control is omission of the primary antibody. While this control addresses whether the secondary antibody reagents are a source of staining, inadvertent binding of the primary antibody to the tissue can occur. Various tissue elements such as Fc receptors and charged molecules may bind the primary antibody non-specifically. Simply omitting the primary antibody as a negative control would miss potential false positive staining by this means. A few “publication worthy” negative controls for IHC are listed below: 1) Preabsorption of primary antibody with the immunogen used to generate the antibody can be employed. The working dilution of the primary antibody and an optimized concentration of the immunogen are incubated together for a period prior to application to the specimen. Lack of staining would indicate specificity of the primary antibody to the target antigen in solution. Positive staining using this method, however, may indicate lack of specificity of the primary antibody and/or the primary antibody is being bound by tissue elements. To rule out the latter, this control can be used in combination with suggestion no. 2 below. Note that adsorption controls are not always feasible or practical depending on the cost or source of the immunogen. 2) Use of an isotype control (e.g. “non-immune” mouse IgG), matched to that of the primary antibody and applied at the same protein concentration as the primary antibody, is probably the most widely used negative control. This control addresses whether tissue elements are inadvertently binding immunoglobulin from the same species as the primary antibody, in addition to non-specific binding from the secondary detection reagents. In most cases, use of a sub-class of isotype immunoglobulin (e.g. mouse IgG2a or IgG2b) is not required. Note that the use of pre-immune immunoglobulin, obtained prior to immunization, could also be used. However, it is very unusual for commercial vendors to offer pre-immune immunoglobulin. 3) Substitution of the primary antibody with an “irrelevant antibody” is also a suitable negative control. The term “irrelevant” refers to a primary antibody of the same isotype as the specific primary antibody (i.e. mouse IgG) and applied at the same concentration, that is known not to bind to a target in the tissue specimen. An example would be an anti-cytokeratin antibody on smooth muscle tissue. As with negative control no. 1 above, lack of staining indicates tissue elements are not binding this isotype of immunoglobulin. 4) In some cases, the target antigen can be removed from the tissue specimen as a sort of “knock-out” preparation. Once the target antigen has been removed, the complete assay is run to determine lack of staining. One method to remove the target antigen is using defined enzyme digestion. Examples include the use of a collagenase if the target antigen is collagen, or hyaluronidase if the target antigen is hyaluronic acid. Of course, there are limitations to this approach, however, variations of this “deletion” or “knock-out” approach would be valid negative controls.

Description

Avidin DCS, Texas Red® labeled, is a cell sorter grade of Texas Red® Avidin D. Through the development of a new method of labeling we are able to incorporate more fluorochrome into a protein without altering the properties of the protein. Not only does the Texas Red® Avidin DCS have a high F/P ratio, the efficiency of fluorescence is also increased. In addition to the improved conjugation technique, we employ a purification step to reduce nonspecific binding which can hamper resolution in cell sorting and other applications.

Description

AMCA Streptavidin is produced by conjugating streptavidin with a coumarin fluorescent dye, 7-amino-4-methylcoumarin-3-acetic acid. This derivative excites in the ultraviolet (350 nm) and emits in the visible (450 nm) producing an intense blue fluorescence.

Amplification of fluorescent signals can be easily achieved with our biotinylated secondary antibodies followed by our highly purified fluorochrome-labeled streptavidin or avidin. Using a biotin/avidin or biotin/streptavidin detection system results in an additional layer of amplification over a directly conjugated secondary antibody.

Technical Information

Vector Laboratories fluorochrome-conjugated streptavidin and avidin reagents are highly purified and possess very low non-specific binding properties. They have extremely high affinity for biotin. These fluorescent conjugates can be used to detect biotinylated secondary antibodies and other macromolecules in applications such as immunofluorescence, in situ hybridization, or flow cytometry.

Most paraffin-embedded tissues have little autofluorescence under those conditions. AMCA Streptavidin may provide an advantage in many labeling situations over conjugates of fluorescein and related dyes. AMCA Streptavidin can be used on frozen or paraffin-embedded sections as a single label or in combination with other fluorescent conjugates for multiple labeling studies.
