Description

Tomato lectin, although sharing some specificities with potato lectin, Datura lectin, and wheat germ agglutinin, has been reported to be dissimilar in many respects. LEL binds well to glycophorin and Tamm-Horsfall glycoprotein and has been used effectively to label vascular endothelium in rodents.

Biotinylated tomato lectin has an appropriate number of biotins bound to provide the optimum staining characteristics for this lectin. This conjugate is supplied essentially free of unconjugated biotins and is preserved with sodium azide.

Can the biotinylated Lycopersicon esculentum (tomato) lectin be used for in vivo perfusion studies to trace blood vessels in mice?

The Lycopersicon esculentum (tomato) lectin has been widely reported as an effective blood vessel marker for in vivo vascular perfusion studies in rodent species. Investigators have primarily utilized one of the fluorophore conjugated tomato lectin formats to trace blood vasculature in normal and diseased animals via tail vein or intracardiac injection. However, the biotinylated format has also been used. It allows for flexibility in subsequent visualization by way of either fluorescence or enzyme-based methods. Published references are best source for procedural details. Examples of references where biotinylated tomato lectins have been applied via in vivo perfusion are featured below: Robertson, R.T., et al. (2014) Histochem. Cell Biol. 143(2) Thurston, G., et al. I recently purchased a biotinylated lectin. The datasheet supplied with the lectin suggests including 0.1 mM Ca++as part of the recommended buffer to prepare a working solution. What should I specifically add, and why is this required?

From our experience we have found that some lectins require Ca++ to be present for optimal binding activity. We suggest using calcium chloride (CaCl2) to fortify working solutions and ensure a minimum level of Ca++ is meet. This may be particularly pertinent if using phosphate based buffers as diluents and storage solutions.

Technical Information

Tomato lectin is a very stable single subunit glycoprotein containing about 50 percent arabinose and galactose and may form multimeric aggregates in solution. 

This biotinylated lectin is an ideal intermediate for examining glycoconjugates using the Biotin-Avidin/Streptavidin System. First the biotinylated lectin is added, followed by the VECTASTAIN® ABC Reagent, Avidin D conjugate, or streptavidin derivative.

Description

Maackia amurensis lectin I binds gal (β-1,4) glcNAc but tolerates substitution of N-acetyllactosamine with sialic acid at the 3 position of galactose. However, MAL I does not appear to bind this structure when substitution with sialic acid is on the 6 position of galactose.

Biotinylated Maackia amurensis lectin has an appropriate number of biotins bound to provide the optimum staining characteristics for this lectin. This conjugate is supplied essentially free of unconjugated biotins and is preserved with sodium azide.

Description

Phaseolus vulgaris agglutinin is the name ascribed to a family of lectins, each of which consists of four subunits. There are two different types of subunits. One appears to be involved primarily in red cell agglutination and has been designated the “E” subunit (for erythroagglutinin). The other type is involved in lymphocyte agglutination and mitogenic activity and has been termed the “L” subunit (for leucoagglutinin). These subunits combine to produce five isolectins.  PHA-L, with four “L” type subunits, does not agglutinate red cell but is a potent mitogen.

Biotinylated PHA-L has an appropriate number of biotins bound to provide the optimum staining characteristics for this lectin. This conjugate is supplied essentially free of unconjugated biotins and is preserved with sodium azide.

PHA-L, with four L type subunits, does not agglutinate red cells but is a potent mitogen. The other three isolectins, designated E3L1, E2L2, and E1L3, have erythroagglutinating and mitogenic activities proportional to the number of respective E or L subunits. We have termed the mixture of the five isolectins PHA (E+L).

This biotinylated lectin is an ideal intermediate for examining glycoconjugates using the Biotin-Avidin/Streptavidin System. First the biotinylated lectin is added, followed by the VECTASTAIN ABC Reagent, Avidin D conjugate, or streptavidin derivative.

 
Note: Do not use a biotinylated PHA-L to perform anterograde neuronal tracing. Only the unconjugated PHA-L (catalog # L-1110) is compatible for this application.

Description

SBA preferentially binds to oligosaccharide structures with terminal α- or β-linked N-acetylgalactosamine, and to a lesser extent, galactose residues. Binding can be blocked by substitutions on penultimate sugars, such as fucose attached to the penultimate galactose in blood group B substance.

Biotinylated Soybean agglutinin has an appropriate number of biotins bound to provide the optimum staining characteristics for this lectin. This conjugate is supplied essentially free of unconjugated biotins and is preserved with sodium azide.

Composed of four subunits of approximately equal size, soybean agglutinin is a family of closely related isolectins. This glycoprotein has a molecular weight of about 120 kDa and an isoelectric point near pH 6.0. 

An important application for SBA is the separation of pluripotent stem cells from human bone marrow. Cells fractionated by SBA do not produce graft vs host disease and can be used in bone marrow transplantation across histocompatibility barriers.

Description

The receptor sugar for WGA is N-acetylglucosamine, with preferential binding to dimers and trimers of this sugar. WGA can bind oligosaccharides containing terminal N-acetylglucosamine or chitobiose, structures which are common to many serum and membrane glycoproteins. Bacterial cell wall peptidoglycans, chitin, cartilage glycosaminoglycans, and glycolipids can also bind WGA.

Wheat germ agglutinin (WGA) contains a group of closely related isolectins, with an isoelectric point about pH 9. Description

Tomato lectin (from Lycopersicon esculentum) is an effective marker of blood vessels and microglial cells in rodents. Conjugation of the lectin with a fluorophore facilitates fast, one-step detection and visualization using intravascular perfusion methods or direct application to tissue sections.

DyLight® 594 labeled tomato lectin has an appropriate number of fluorochromes bound to provide the optimum staining characteristics for this lectin. This conjugate is supplied essentially free of unconjugated fluorochromes.

Can the biotinylated Lycopersicon esculentum (tomato) lectin be used for in vivo perfusion studies to trace blood vessels in mice?

The Lycopersicon esculentum (tomato) lectin has been widely reported as an effective blood vessel marker for in vivo vascular perfusion studies in rodent species. Investigators have primarily utilized one of the fluorophore conjugated tomato lectin formats to trace blood vasculature in normal and diseased animals via tail vein or intracardiac injection. However, the biotinylated format has also been used. It allows for flexibility in subsequent visualization by way of either fluorescence or enzyme-based methods. Published references are best source for procedural details. Technical Information

Tomato lectin is a very stable single subunit glycoprotein containing about 50 percent arabinose and galactose and may form multimeric aggregates in solution. Tomato lectin, although sharing some specificities with potato lectin, Datura lectin, and wheat germ agglutinin, has been reported to be dissimilar in many respects. LEL binds well to glycophorin and Tamm-Horsfall glycoprotein and has been used effectively to label vascular endothelium in rodents.

Texas Red® or DyLight® 594 conjugated tomato lectin provides an excellent contrast to green/yellow fluorescence such as GFP expressed in transgenic animals, or with fluorescein conjugates in standard double label studies. The tomato lectin complements our existing range of lectin reagents and should be a valuable tool in examining rodent tumor angiogenesis, tracing neovascular development in xenograft models and brain research.

Accompanying each fluorescent lectin is an analysis data sheet summarizing the results of our quality control tests and providing pertinent information on the product. All of these reagents are supplied as solutions preserved with sodium azide.

Description

Unlike Ulex europaeus and Lotus tetragonolobus lectins which prefer (α -1,2) linked fucose residues, Aleuria aurantia lectin binds preferentially to fucose linked (α -1,6) to N-acetylglucosamine or to fucose linked (α -1,3) to N-acetyllactosamine related structures. AAL also reversibly binds fucose attached to nucleic acids.

Fluorescein labeled Aleuria aurantia lectin has an appropriate number of fluorochromes bound to provide the optimum staining characteristics for this lectin. This conjugate is supplied essentially free of unconjugated fluorochromes. The excitation maximum is at 495 nm and the emission maximum is at 515 nm.

Description

The carbohydrate binding site recognizes (β-1,4) linked N-acetylglucosamine oligomers, preferring chitobiose or chitotriose over a single N-acetylglucosamine residue. This lectin binds well in the acidic pH range but its affinity decreases above pH 8.0.

DSL also binds well to N-acetyllactosamine and oligomers containing repeating N-acetyllactosamine sequences. A branched pentasaccharide including two N-acetyllactosamine disaccharides linked to mannose (β-1,6) and (β-1,2) was reported to be the most potent inhibitor of agglutination. 

Fluorescein labeled DSL has an appropriate number of fluorochromes bound to provide the optimum staining characteristics for this lectin. This conjugate is supplied essentially free of unconjugated fluorochromes. The excitation maximum is at 495 nm and the emission maximum is at 515 nm.

Description

VVL recognizes preferentially α- or β-linked terminal N-acetylgalactosamine, especially a single α-N-acetylgalactosamine residue linked to serine or threonine in a polypeptide (the Tn antigen). Evidence suggests that this lectin also may require specific amino acid sequences at the receptor site of glycosylation. The disaccharide galactosyl (α-1,3) N-acetylgalactosamine is also a potent inhibitor of this lectin.
