Technical Information

All the reagents in the VectaFluor Excel Kit are affinity purified and extensively cross-adsorbed to ensure high sensitivity and low background. In addition, the VectaFluor Reagents are conjugated to the DyLight dyes using technology that ensures maximum labeling without compromising antibody affinity or specificity. DyLight dyes offer several advantages such as brighter fluorescence, greater photostability, and pH independence.

All reagents are supplied in stabilized, R.T.U. solutions for maximum convenience.

Description

Fluorescein Goat Anti-Human IgE, epsilon chain specific, can be used for immunofluorescence and other applications. Optimal F/P ratios have been established for each conjugate to ensure maximum fluorescence with minimal background staining.

Technical Information

The goat anti-human Ig antibodies are prepared by hyperimmunizing animals in a manner that produces high affinity antibodies. These are then purified by an affinity chromatography procedure designed to remove any low affinity antibodies which may be present. Cross-reactivities that are likely to interfere with specific labeling are removed by solid-phase adsorption techniques. The final product is then subjected to rigorous quality control assays including immunodiffusion, solid-phase enzyme immunoassays, gel electrophoresis and solid-phase binding assays. In preparing the labeled antibodies, great care is taken to ensure the maximum degree of labeling with no alteration in the specificity and affinity of the antibody. The labeled antibody then undergoes a further series of quality control assays, including immunohistochemical analysis.

Description

Avidin DCS, Fluorescein labeled, is a cell sorter grade of Fluorescein Avidin D. Through the development of a new method of fluorescein labeling we are able to incorporate more fluorochrome into a protein without altering the properties of the protein. Not only does the Fluorescein Avidin DCS have an optimal F/P ratio, the efficiency of fluorescence is also increased. In addition to the improved conjugation technique, we employ a purification step to reduce nonspecific binding which can hamper resolution in cell sorting and other applications.

Technical Information

Vector Laboratories fluorochrome-conjugated streptavidin and avidin reagents are highly purified and possess very low non-specific binding properties. They have extremely high affinity for biotin. Amplification of fluorescent signals can be easily achieved with our biotinylated secondary antibodies followed by our highly purified fluorochrome-labeled streptavidin or avidin. Using a biotin/avidin or biotin/streptavidin detection system results in an additional layer of amplification over a directly conjugated secondary antibody.

Since the excitation and emission maxima are well separated from those of fluorescein, Texas Red® Streptavidin can be employed with Fluorescein Streptavidin or other fluorescein conjugates to simultaneously localize two antigens in the same tissue section. Texas Red® Streptavidin is also ideal for flow cytometry using instruments equipped with dye lasers.

Description

These antibodies to lectins are produced by immunizing goats with pure lectins. Following conventional purification steps, specific antibody is isolated by affinity chromatography on lectin-agarose columns. This anti-lectin is supplied in solution. These highly purified specific antibodies provide excellent intermediate reagents for localizing lectin receptors in tissues and on cells or following lectin transport in neuronal tracing.

Description

This lectin appears to bind only O-glycosidically linked oligosaccharides, preferring the structure galactosyl (β-1,3) N-acetylgalactosamine. This structure (the T-antigen) is the oligosaccharide to which peanut agglutinin (PNA) binds. However, unlike PNA, Jacalin will bind a mono- or disialylated form of this structure. This lectin has been used to purify human IgA. The specificity of this lectin also affords the opportunity to localize or isolate glycoproteins with O-glycosidically linked oligosaccharide side chains.

Biotinylated Jacalin has an appropriate number of biotins bound to provide the optimum staining characteristics for this lectin. This conjugate is supplied essentially free of unconjugated biotins and is preserved with sodium azide.

For most applications we recommend a freshly prepared working solution of 5-20 µg/ml in the above buffer.

I recently purchased a biotinylated lectin. The datasheet supplied with the lectin suggests including 0.1 mM Ca++as part of the recommended buffer to prepare a working solution. What should I specifically add, and why is this required?

From our experience we have found that some lectins require Ca++ to be present for optimal binding activity. We suggest using calcium chloride (CaCl2) to fortify working solutions and ensure a minimum level of Ca++ is meet. This may be particularly pertinent if using phosphate based buffers as diluents and storage solutions.

Description

Lotus tetragonolobus lectin is a family of closely related glycoproteins that appear to have similar specificities toward α-linked L-fucose containing oligosaccharides. Although many of the binding properties of Lotus lectin are similar to those of Ulex europaeus lectin I, the binding affinities and some specificities for oligosaccharides are significantly different between these fucose-specific lectins.

Biotinylated Lotus tetragonolobus has an appropriate number of biotins bound to provide the optimum staining characteristics for this lectin. This conjugate is supplied essentially free of unconjugated biotins and is preserved with sodium azide.

Description

NPL, isolated from daffodil bulbs, has a specificity toward α-linked mannose, preferring polymannose structures containing (α-1,6) linkages. Binding to mannose polymers can occur via internal mannose residues and is not dependent on structural integrity of a non-reducing end sugar. NPL also binds some galactomannans, and differs in other binding characteristics from a related lectin, Galanthus nivalis lectin. Unlike Con A, LCA, or PSA, NPL does not bind glucose.

Biotinylated Narcissus pseudonarcissus lectin has an appropriate number of biotins bound to provide the optimum staining characteristics for this lectin. This conjugate is supplied essentially free of unconjugated biotins and is preserved with sodium azide.
