核酸清除剂, 本品使用不需要分离管, 即可去除低分子DNA片断(≤50bp)

DNA Cleaner 本品不需要使用有机溶剂或离心管，35分钟内即可高效清除50bp 以下DNA 片断。另外，本品还可用于dsRNA 的纯化。最适合用于PCR 后的引物、引物对、dNTP、DNA 聚合酶的清除和限制性内切酶处理后的酶消除。

同时，利用乙醇沉淀法将ABI 公司BigDye Cycle Sequencing kit 扩增序列的DNA 样品纯化时，使用本品能得到清晰的序列结果。

	DNA Cleaner	Q公司柱色谱法
Step 1	添加与DNA溶液等量的DNA Cleaner。	向PCR样品中添加5倍容量的结合液。此时，若混合物的颜色不为黄色，请添加3M醋酸钠。
Step 2	在室温下静置10分钟。	向离心管中添加Step 1.的样品。
Step 3	在室温下离心分离10分钟。	在室温下离心分离1分钟。
Step 4	除去上清液。添加70%乙醇。	除去滤液。添加洗涤液。
Step 5	在室温下离心分离1分钟。	在室温下离心分离1分钟。
Step 6	上清除去。干燥5分钟。	除去滤液。
Step 7	TE Buffer、灭菌水溶解	在室温下离心分离10分钟。
Step 8	FINISH	添加50μL析出液。静置1分钟。

原理

1. 用α-淀粉酶、蛋白酶和淀粉糖苷酶，通过添加四倍体积的95%乙醇沉淀未分解的残余大分子，然后用含硅藻土的玻璃过滤坩埚捕获。
2. 总膳食纤维含量是通过从玻璃过滤坩埚上捕获的残渣的干重中扣除蛋白质和灰分的含量来确定的。
3. 1. 可溶性膳食纤维和不溶性膳食纤维含量的分数分析

Principle

   2. Digestible components in samples, such as starch and protein, are hydrolyzed
   3. with alpha-amylase, protease, and amyloglucosidase, precipitate undecomposed residual macromolecule by adding quadruple volume 95% Ethanol, and then trapped with a glass filter crucible containing diatomaceous earth.
   4. Total Dietary Fiber Contents are determined by deducting weight of protein and ash content from the dry weight of trapped residue on a glass filter crucible.
   5. 1. Fractional Analysis of Soluble Dietary Fiber Contents and Insoluble Dietary Fiber Contents
   6. Digestible components in samples, such as starch and protein, are hydrolyzed with alpha-amylase, protease, and amyloglucosidase, then filtrated with a glass filter crucible containing diatomaceous earth.
   7. Insoluble Dietary Fiber Contents are determined by deducting weight of protein and ash content from the dry weight of trapped insoluble residue on a glass filter crucible.
   8. Meanwhile, add a quadruple volume 95% Ethanol into the filtrate, precipitate undecomposed residual soluble macromolecule, and trap with a glass filter crucible containing diatomaceous earth. Soluble Dietary Fiber Contents are determined by deducting weight of protein and ash content from the dry weight of the trapped precipitated residue on a glass filter crucible.
   9. Total Dietary Fiber Contents are defined as the sum of Insoluble Dietary Fiber Contents and Soluble Dietary Fiber Contents.
   10. 1. Determination of Low-molecular Soluble Dietary Fiber
   11. Specific Low-molecular Soluble Dietary Fibers, which are not precipitated even by addition of a quadruple volume of 95% Ethanol, are determined by high-performance liquid chromatography.
   12. After digestible components such as starch and protein in samples are hydrolyzed with alpha-amylase, protease, and amyloglucosidase, the undecomposed residual macromolecule are precipitated by adding a quadruple volume of 95% Ethanol. Then filter it with a glass filter crucible containing diatomaceous earth.
   13. Proteins, organic acids and inorganic salts in the filtrate are removed with ion-exchange resin, and then the solution is applied to high-performance liquid chromatography. Low-molecular Soluble Dietary Fiber Content is determined by the ratio of the peak area of the sample with that of glucose (internal standard) on the chromatogram.
   14. Meanwhile, Insoluble and Soluble Macromolecule Dietary Fiber Content is determined by deducting weight of protein and ash content from the dry weight of the trapped precipitated residue on a glass filter crucible.

Total Dietary Fiber Contents is defined as sum of Low-molecular Soluble Dietary Fiber Contents and Insoluble and Soluble Macromolecular Dietary Fiber Contents.
